"A loxP site was inserted 40 nucleotides 5' of the initiation methionine codon in exon 3 and a floxed neomycin resistance gene as well as an EGFP gene were inserted in the intron between exons 5 and 6. The EGFP insert was non functional. Cre mediated recombination removed the neo cassette leaving the loxP sites in exon 3 and intron 5 intact."

"Homozygotes exhibit no overt phenotype and that the SK1-flox (delta neo) allele may confer a null phenotype even before exposure to Cre recombinase: RT-PCR shows the EGFP sequence disrupts splicing such that no full length transcripts are detected. No EGFP expression is reported. When bred to mice that express Cre recombinase, the SK1 sequences encoding the translation initiation site through transmembrane domain five are deleted in the cre-expressing tissues of the offspring."
